Bacteria don't have TATA boxes since TBP is a eukaryotic/archaeal protein.

Probably not. They would need a host cell which would, at the very least, need to be related to the species they're trying to create. The paper describes taking a bacterium, removing its own DNA and replacing it with DNA that has been made artificially. Without a home, i.e. the host cell, the DNA is useless.
